黄斑明目颗粒对N-甲基-N-亚硝脲诱导视网膜变性大鼠的保护作用

摘要:
目的 观察黄斑明目颗粒对N-甲基-N-亚硝脲(MNU)诱导视网膜变性大鼠的保护作用。方法 生后43 d的雌性SD大鼠30只随机分为3组(n=10):正常对照组、模型对照组、黄斑明目颗粒组。黄斑明目颗粒组以黄斑明目颗粒溶解后灌胃,正常对照组和模型对照组以等量蒸馏水灌胃,每日2次,连续7 d。于生后50 d,黄斑明目颗粒组和模型对照组大鼠腹腔注射MNU 40 mg·kg-1,正常对照组注射等量生理盐水。观察MNU处理后12,24,48,72 h各组大鼠视网膜电图中a波及b波的变化,并于MNU处理后72 h处死大鼠,取眼球做组织学观察。结果 正常对照组大鼠不同时间点视网膜电图(ERG)a波及b波的振幅差异无显著性的意义,模型对照组在MNU处理后ERG a波及b波的振幅均呈持续性、进行性下降,黄斑明目颗粒可明显抑制ERG a波及b波振幅的降低。通过视网膜透视电子显微镜的组织学观察显示,黄斑明目颗粒组的病理组织学损害较模型对照组明显减轻。结论 黄斑明目颗粒对MNU诱导的视网膜变性大鼠有保护作用,是治疗视网膜变性疾病的有效中药复方制剂。

Abstract:
OBJECTIVE To observe the effects of Huangbanmingmu granule on N-methyl-N-nitrosourea (MNU) induced retinal degeneration in SD rats. METHODS Forty-three-day female SD rats were randomly divided into three groups(n=10): control group, model group, Huangbanmingmu granule group. Huangbanmingmu granule were irrigated into rats in Huangbanmingmu granule group for 7 days twice a day. Control group and model group were given equal water. Fifty-day SD rats received a single intraperitoneal injection of 40 mg·kg-1 MNU in model group and Huangbanmingmu granule group, while control group was given equal normal saline. Change of electroretinogram a wave and b wave were observed in 12, 24, 48, 72 hours after MNU injection. The rats were killed in 72 hours after MNU injection and histopathologic changes were observed with transmission electron microscopy. RESULTS The difference of the average amplitude of ERG a wave and b wave was not significant between different time in control group. The average amplitude of ERG a wave and b wave gradually decreased after MNU treatment in model group. Huangbanmingmu granule could evidently inhibit lose of a wave and b wave. The histopathologic injury of rat retina in Huangbanmingmu granule group was lighter than in model control group by means of transmission electron microscopy. CONCLUSION Huangbanmingmu granule has protective effects on MNU induced retinal degeneration in SD rats, and is an effective traditional Chinese compound preparation to treat the diseases of retinal degeneration.
