灰色关联分析中药蒲公英不同部位对RAW264.7细胞的抑制作用

摘要:
目的 研究蒲公英不同部位化学成分的差异及其对RAW264.7细胞的抑制作用区别,探讨蒲公英不同部位的精准应用。方法 使用HPLC建立不同部位的液相图谱并进行成分指认,使用MTT法测定不同部位提取物对炎症细胞RAW 264.7增殖的抑制作用,使用灰色关联软件,将色谱峰的峰面积与抗炎药效相关联,计算每个成分的贡献率。结果 蒲公英根、茎、叶、花中的成分种类与含量均不同。在体外药效研究中,蒲公英花的效果最好,对炎症细胞RAW264.7的抑制率达到65.51%。蒲公英根、叶次之,蒲公英茎对炎症细胞的抑制作用不大。灰色关联分析结果显示,绿原酸、橙皮苷、香叶木素、木犀草素、异鼠李素对抑制炎症细胞增殖的贡献程度与其他成分相比存在较大差异。结论 蒲公英不同部位的体外药效存在差异,本实验明确了蒲公英的精准药用部位。

Abstract:
OBJECTIVE To Study on the difference of chemical constituents in different parts of Taraxaci Herba and the inhibitory effect to the RAW264.7 cells which aim to explore the precision use of Taraxaci Herba. METHODS The liquid phase chromatogram of different parts was established by using HPLC and identified with components, the inhibitory effect of different parts extracts on the proliferation of inflammatory cell RAW264.7 was determined by using MTT method, and used gray association software to associate the peak area of the chromatographic peak with the anti-inflammatory drug effect to calculate the contribution rate of each component. RESULTS Different species and contents of components in the roots, stems, leaves, and flowers were different in Taraxaci Herba. In vitro efficacy studies, the flowers of Taraxaci Herba worked best effect on inhibition of RAW264.7 cells which the inhibition rate was 65.51%. The leaves and roots of Taraxaci Herba ranked second. The results of grey relational analysis showed that the chlorogenic acid, hesperidin, diosmetin, luteolin, isorhamnetin had greater degrees of contribution on inhibition RAW264.7 cells was quite different compared with other components. CONCLUSION There are differences in different parts of Taraxaci Herba. This experiment clarified the precise medicinal site of Taraxaci Herba.
