中压制备色谱法制备高纯度石杉碱甲

摘要:
目的 采用中压制备色谱法从蛇足石杉浸膏中制备高纯度石杉碱甲。方法 分别使用聚合物反相填料和C18反相填料为分离介质,串联两步中压制备色谱,从蛇足石杉浸膏中分离石杉碱甲单体。结果 制备的石杉碱甲单体纯度为99.7%,总体回收率达到87.5%。结论 该法利用聚合物填料和C18填料的各自特点,通过合理的工艺衔接,为获得医药级石杉碱甲提供了一条高效、绿色和经济的途径。

Separation and Preparation of high-purity huperzine A by medium pressure preparative chromatography

Abstract:
OBJECTIVE To establish a method for the preparation of high-purity huperzine A from the extract of huperzia serrata. METHODS By using the polymer reversed phase medium and C18 reversed phase medium and two steps of medium pressure preparation in series, huperzine A monomer was isolated and prepared from the huperzia serrata extract. RESULTS The purity of 99.7% huperzine A monomer was obtained, and the overall recovery rate was 87.5%. CONCLUSION This method utilizes the characteristics of polymer medium and C18 medium to provide an efficient, green and economical way to obtain pharmaceutical grade huperzine A through technological linkage.
