酶法协同超声提取防己总生物碱的工艺研究 |

摘要: |
目的 优选酶法协同超声提取防己总生物碱的工艺条件。方法 以总生物碱的提取率为研究指标,在单因素试验的基础上,通过正交试验和响应面试验,对酶法辅助超声波提取防己总生物碱的工艺条件进行优化。结果 确定最佳工艺条件:药材粉末加2%的复合酶在pH 4.5,50℃的条件下酶解1 h,然后用17倍量的74%乙醇超声提取2次,每次30 min,超声功率为165 W,超声温度为40℃。在此条件下,防己总生物碱的提取率可达3.92%。结论 酶法协同超声提取防己总生物碱是一种有效的提取方法,该法操作简便、结果可靠,适合于工业化大生产。 |

OBJECTIVE To optimize extraction method of total alkaloids from Stephania tetrandra by enzyme with ultrasonic technology. METHODS Orthogonal experiment and response surface methodology was employed to optimize the technology conditions for extracting total alkaloids from Stephania tetrandra on the basis of single factor test. Taking total alkaloids extraction yield as evaluation index, optimize extraction conditions of total alkaloids from Stephania tetrandrs by enzyme with ultrasonic technology. RESULTS The optimum condition were as follows:compound enzyme was 2%, enzyme solution pH was 4.5, enzymolysis time was 1 h, enzyme solution temperature was 50℃, use 17 times of 74% ethanol ultrasound 30 min for 2 times, ultrasonic power was 165 W, ultrasonic temperature was 40℃. Under these conditions, the extraction rate of total alkaloids from Stephania tetrandra was 3.92%. CONCLUSION Ultrasonic assisted enzymatic extraction total alkaloid from Stephania tetrandra is an effective extraction method. The extraction technology is simply, stable, feasible and suitable for large industrial production. |
