S-腺苷蛋氨酸对子宫平滑肌收缩的影响 |

摘要: |
目的 探究S-腺苷蛋氨酸对子宫平滑肌收缩的影响,为寻找安全有效的新型子宫收缩调节药物提供理论依据。方法 取未妊娠、妊娠早期(6.5 d)、妊娠中期(12.5~13.5 d)和妊娠晚期(16.5~17.5 d)ICR小鼠,分离子宫,制备成1 cm长子宫肌条,两端分别固定于张力换能器,采用氨甲酰胆碱诱导收缩,累积给药法给予沙丁胺醇、S-腺苷蛋氨酸、蛋氨酸、牛磺酸等药物,记录、测量并分析离体子宫收缩曲线。结果 沙丁胺醇和S-腺苷蛋氨酸对未孕、妊娠早期(6.5 d)、妊娠中期(12.5~13.5 d)和妊娠晚期(16.5~17.5 d)4个时间点的小鼠离体子宫平滑肌的收缩均有较强的抑制作用,分别在10-7 mol·L-1和1.0 mmol·L-1的溶液浓度下显著抑制收缩。蛋氨酸、牛磺酸(10 mmol·L-1)在实验浓度下对ICR小鼠离体子宫平滑肌的收缩影响不明显。结论 S-腺苷蛋氨酸可有效抑制不同阶段离体子宫的收缩,提示S-腺苷蛋氨酸可能是一个有效且安全的子宫收缩调节药物。 |

OBJECTIVE To investigate the effect of S-adenosylmethionine(SAMe) on contraction of uterine smooth muscle, and to provide a theoretical basis for novel drug for preterm labor with good characters in safety and effectiveness. METHODS The uterus was dissected from mice at the different time-points: non-pregnant, and earlypregnancy (6.5 d), mid pregnancy (12.5-13.5 d) and late pregnancy (16.5-17.5 d) and the uterus was divided into about 1 cm length in the direction of the longitudinal muscle fibers in the Lock's solution. The muscle strips were fixed to the transducers at both ends. Sulbactamol, SAMe, methionine and taurine were administered by the cumulative concentration administration method, the uterine contraction curves were recorded, measured and analyzed. RESULTS The strong inhibitory effects of salbutamol and SAMe on contraction of isolated uterine smooth muscle of the mice in the early stage (6.5 d), the second trimester (12.5-13.5 d) and the third trimester (16.5-17.5 d) were observed. Salbutamol 10-7 mol·L-1 and SAMe 1.0 mmol·L-1 signifantly inhibited contraction. The effects of the other amino acids (methionine and taurine) on the contraction of isolated uterine smooth muscle in mice were not significant. CONCLUSION SAMe can effectively inhibit the contraction of uterine phases of pregnancy, so SAMe may be a novel relaxation drug for uterine smooth muscle. |
