目的 探讨畲药山里黄根水提物对卡介苗(bacillus calmette-guérin,BCG)和脂多糖(lipopolysaccharide,LPS)诱导的小鼠免疫性肝损伤的保护作用。方法 ICR小鼠,♂,随机分为正常组、模型组、山里黄根高、中、低剂量组和联苯双酯组。小鼠尾静脉注射BCG和LPS诱导小鼠免疫性肝损伤,正常组注射等体积的生理盐水。全自动生化分析仪分析小鼠血清中丙氨酸转氨酶(ALT)、天门冬氨酸氨基转移酶(AST)活性,ELISA法测定血清中IL-2、IL-4、IL-10和TNF-γ的含量。HE染色观察小鼠肝脏的病理改变。结果 与模型组相比,山里黄根水提物能显著降低小鼠血清ALT、AST的活性(P<0.05),显著升高炎性因子IL-2与IL-10水平(P<0.05),显著降低炎性因子IL-4与TNF-γ水平(P<0.05)。与模型组相比,山里黄根水提物组小鼠肝组织炎症细胞浸润、水肿、坏死结构和免疫复合沉淀物均明显减轻。结论 山里黄根水提物对BCG联合LPS诱导的免疫性肝损伤具有保护作用。 |

OBJECTIVE To explore the protective effect of She national medicine (root of Gardenia jasminoides extraction) on bacillus calmette-guérin(BCG) and lipopolysaccharide(LPS)-induced immunological liver injury in mice. METHODS ICR male mice were randomly divided into normal group, model group, the water extract of high, medium and low dose group and bifendate group. Immunological liver injury model were established by intravenously injection of BCG and LPS. The mice in normal group were injected of normal saline. The activity of ALT and AST activity in serum of mice were detected by full automatic biochemical analyzer. ELISA method was used to measure the content of serum IL-2, IL-4, IL-10, TNF-γ. HE staining was used to observe the pathological changes of the liver in mice. RESULTS Compared with the model group, the aqueous extract significantly reduced the activity of ALT and AST(P<0.05); significantly increased the level of IL-2 and IL-10 (P<0.05); and reduced the level of IL-4 and TNF-γ(P<0.05). Compared with the model group, the inflammatory cells in the aqueous extract groups mice liver tissue infiltration, edema, necrosis and immune complex deposit structure were obviously alleviated. CONCLUSION The aqueous extract of She medicine-root of Gardenia jasminoides has protective effect on immune hepatic injury induced by BCG combined with LPS. |
