毛细管微萃取结合微顺序注射在线检测全血中异丙酚含量

摘要:
目的 建立一种快速测定全血中异丙酚含量的原位、在线分析的新方法。方法 利用毛细管微萃取器联合紫外光纤传感仪及微顺序注射-阀上实验室系统在线测定全血中异丙酚含量。结果 该方法的加样回收率为95.8%~103.6%;低、中、高 3 个浓度(100,150,200 μg·mL-1)的日内精密度RSD分别为7.2%,8.2%和2.6%,日间精密度RSD分别为10.0%,6.1%和7.7%。与传统液液萃取方法相比,该方法异丙酚平均萃取率提高3.5%,萃取时间缩短5~7 min。结论 该方法样品前处理无需离心、除蛋白等操作, 能快速测定全血中异丙酚的含量, 为全血中异丙酚含量原位、在线检测提供了良好的手段。

Online Determination of Propofol in Whole Blood by Combining Capillary Micro-extraction with Micro-sequential Injection

Abstract:
OBJECTIVE To determine the propofol’s content in whole blood online and in situ by combined capillary micro-extraction rapid extraction with optical-fiber. METHODS Capillary micro-extractor, optical-fiber sensing devices and microSIA lab-on-valve were used to determine the propofol in whole blood. RESULTS The new methods of recovery ranged in 95.8%-103.6%. The intra-day RSDs were 7.2%, 8.2% and 2.6%, and inter-day RSDs were 10.0%, 6.1% and 7.7%, respectively at 3 concentrations(100, 150, 200 μg·mL-1). The?average?extraction?rate was higher than tradition liquid-liquid extraction about 3.5%, and the extraction time was still shorter about 5-7 min. CONCLUSION This new method could test propofol in whole blood online and in situ without using centrifuge and removing protein, and provide an excellent opition to determine?the?propofol?in?whole?blood.
