瑞香素对三阴性乳腺癌细胞株MDA-MB-231的抑制作用及其机制研究

摘要:
目的 探讨瑞香素对三阴性乳腺癌细胞株MDA-MB-231的增殖、迁移及侵袭的抑制作用及其潜在机制。方法 体外培养MDA-MB-231细胞,随机分为对照组和瑞香素10,20,40 μg·mL–1组。以MTT法检测细胞的增殖情况;通过克隆形成试验检测各组MDA-MB-231细胞克隆形成情况;划痕试验及Transwell观测细胞迁移和侵袭情况;Western blotting检测Vimentin、MMP9、Cyclin D1、CDK4蛋白表达量。结果 与对照组相比,瑞香素组(10,20,40 μg·mL–1)显著抑制细胞增殖(P<0.05或P<0.01);瑞香素组(10,20,40 μg·mL–1)可以抑制人乳腺癌细胞株MDA-MB-231的克隆形成(P<0.01);瑞香素组(20,40 μg·mL–1)细胞的迁移能力和侵袭能力显著下降(P<0.01);瑞香素组(20,40 μg·mL–1)细胞Vimentin、MMP9、Cyclin D1、CDK4蛋白表达量显著降低(P<0.01)。结论 瑞香素可以抑制三阴性乳腺癌细胞株MDA-MB-231的增殖、迁移及侵袭能力,其作用机制可能与下调Vimentin、MMP9、Cyclin D1、CDK4的表达有关。

Abstract:
OBJECTIVE To explore the inhibitory effect of daphnetin on proliferation, migration and invasion of human triple negative breast cancer cell MDA-MB-231 and its potential mechanism. METHODS Mda-mb-231 cells were cultured in vitro and randomly divided into control group and daphnetin 10, 20, 40 μg·ml-1 groups. MTT assay was used to detect the MDA-MB-231 cells proliferation. The clone formation of MDA-MB-231 cells in each group was detected by clone formation experiment. Scratch and Transwell assays were used to observe the migration and invasion of cells. Western blotting was used to detect expression of Vimentin, MMP9, Cyclin D1, and CDK4 proteins. RESULTS Compared with the control group, daphnetin groups(10, 20, 40 μg·ml-1) significantly inhibited the proliferation of MDA-MB-231 cells(P<0.05 or P<0.01), daphnetin groups(10, 20, 40 μg·ml-1) could inhibit the colony formation of human breast cancer cells MDA-MB-231(P<0.01), the migration and invasion ability of the cells in the daphnetin group(20, 40 μg·ml-1) decreased remarkably compared with the control group(P<0.01), the protein expression levels of Vimentin, MMP9, Cyclin D1 and CDK4 in the daphnetin group(20, 40 μg·ml-1) were significantly reduced(P<0.01). CONCLUSION Daphnetin can inhibit the proliferation, migration and invasion of human breast cancer cell MDA-MB-231, and its mechanism may be related to the down-regulation of the expression of Vimentin, MMP9, Cyclin D1 and CDK4.
