HPLC波长切换法同时测定通幽润燥丸中9种活性成分

摘要:
目的 采用HPLC波长切换法同时测定通幽润燥丸中柚皮苷、新橙皮苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、和厚朴酚、厚朴酚和大黄酚9种活性成分的含量。方法 采用依利特Hypersil OSD2 C18色谱柱(250 mm×4.6 mm,5 μm);以乙腈(A)-0.1%磷酸溶液(B)为流动相进行梯度洗脱;流速为1.0 mL·min-1;柱温为30℃;检测波长为275 m(柚皮苷、新橙皮苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素)和220 m(和厚朴酚、厚朴酚、大黄酚);进样量10 μL。结果 9种活性成分的分离度较好。柚皮苷、新橙皮苷、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、和厚朴酚、厚朴酚和大黄酚进样量分别在0.76~22.94,0.47~14.21,1.46~43.66,0.24~7.11,0.51~15.36,0.13~3.93,0.053~1.58,0.30~8.95,0.38~11.38 mg·mL-1内呈良好线性关系,平均加样回收率(n=6)分别为99.27%,100.04%,98.90%,100.10%,99.67%,98.75%,99.88%,99.59%,99.92%,RSD分别为0.12%,0.15%,0.41%,0.18%,0.28%,0.27%,0.65%,0.46%,0.24%。结论 通幽润燥丸中9种成分地含量测定方法准确,灵敏度高,分离效果好,能较好地评价该品种的质量,为该药物全面的质量控制提供了科学依据。

Abstract:
OBJECTIVE To determine the content of naringin, neohesperidin, baicalin, wogonoside, baicalein, wogonin, honokiol, magnolol and chrysophanol in Tongyou Runzao pills by HPLC wavelength switching method. METHODS The column of illite Hypersil OSD2 C18(250 mm×4.6 mm, 5 μm) was used; the mobile phase of acetonitril(A)-0.1% phosphoric acid solution(B) was used for gradient elution; the flow rate was 1.0 mL·min-1; the column temperature was 30℃; the detection wavelength was 275 nm(naringin, neohesperidin, baicalin, wogonoside, baicalein, wogonin) and 220 nm(honokiol, magnolol, chrysophanol); the injection volume was 10 μL. RESULTS The separation of the nine active constituents was good. The linear range was 0.76-22.94, 0.47-14.21, 1.46-43.66, 0.24-7.11, 0.51-15.36, 0.13-3.93, 0.053-1.58, 0.30-8.95, 0.38-11.38 mg·mL-1for naringin, neohesperidin, baicalin, wogonoside, baicalein, wogonin, honokiol, magnolol and chrysophanol, respectively. The average recoveries(n=6) were 99.27%, 100.04%, 98.90%, 100.10%, 99.67%, 98.75%, 99.88%, 99.59%, 99.92%, respectively with RSDs of the recoveries were 0.12%, 0.15%, 0.41%, 0.18%, 0.28%, 0.27%, 0.65%, 0.46%, 0.24%, respectively.CONCLUSION The method for the determination of 9 components in Tongyou Runzao pills is accurate, sensitive and has good separation effect. It can better evaluate the quality of the drug and provide a scientific basis for the overall quality control of drug.
