光裸方格星虫纤溶酶对FeCl3诱导大鼠颈动脉血栓的抗栓作用研究

摘要:
目的 研究光裸方格星虫纤溶酶(fibrinolytic enzyme from Sipunculus nudus,SNFE)对FeCl3诱导大鼠颈动脉血栓的抗栓作用。方法 SD大鼠随机分为假手术组、模型组、阿司匹林组(100 mg·kg-1)和SNFE高、低剂量组(5 000,2 500 IU·kg-1),连续灌胃给药14 d后,以30% FeCl3诱导颈动脉血栓。成栓1 h后腹主动脉取血,测定凝血4项、组织纤溶酶原激活物(tissue-type plasminogen activator,t-PA)及I型纤溶酶原激活物抑制因子(plasminogen activator inhibitor-I,PAI-I);称量各组血栓的湿重和干重,计算血栓形成抑制率,截取血栓环,进行HE染色。结果 与假手术组比较,模型组凝血酶原时间(prothrombin time,PT)、凝血酶时间(thrombin time,TT)明显缩短,纤维蛋白原(fibrinogen,FIB)含量明显增加(P<0.05或P<0.01);与模型组相比,SNFE高剂量组PT、活化部分凝血酶时间显著延长,FIB含量减少(P<0.05或P<0.01),低剂量组各项指标有改善趋势。与假手术组比较,模型组t-PA和t-PA/PAI-I明显减少(P<0.01),PAI-I无明显差异;与模型组比较,SNFE高、低剂量组的t-PA及高剂量组的t-PA/PAI-I显著增加(P<0.05或P<0.01),PAI-I无明显差异。与模型组比较,SNFE各剂量组的血栓干重和湿重均降低,SNFE高剂量组的干、湿重分别抑制56.86%和41.67%,低剂量组的干、湿重分别抑制21.57%和11.90%。模型组血管腔内有致密血栓,SNFE低剂量组血栓较模型组少,阿司匹林组及高剂量组无明显血栓块。结论 方格星虫纤溶酶可通过影响凝血系统和纤溶系统抑制FeCl3诱导的大鼠颈总动脉血栓的形成。

Sandy on Antithrombotic Effect of Fibrinolytic Enzyme from Sipunculus Nudus on the Carotid Artery Thrombus Induced by FeCl3 in Rats

Abstract:
OBJECTIVE To investigate the antithrombotic effect of fibrinolytic enzyme from Sipunculus nudus(SNFE) on the carotid artery thrombosis induced by FeCl3 in rats. METHODS SD rats were divided into sham surgery group, model group, aspirin group(100 mg·kg-1) and the high and low dose of SNFE groups(5 000 and 2 500 IU·kg-1). After 14 d of continuous intragastric administration, arterial thrombus was induced by 30% FeCl3. Blood samples were collected through aorta abdominals at 1 h after the arterial thrombus formation, and the four coagulation indicators, the plasma levels of tissue-type plasminogen activator(t-PA) and plasminogen activator inhibitor-I(PAI-I) were measured. Thrombus weight (wet and dry weight) in each group was weighed, thrombus formation rate was calculated, and the morphology of thrombus was observed by HE staining. RESULTS Compared with the sham surgery group, the prothrombin time(PT), thrombin time(TT) of the model group was significantly shortened and the content of fibrinogen(FIB) was significantly increased(P<0.05 or P<0.01). Compared with the model group, the prolongation of PT and active partial thromboplastin time, and the decreasing of FIB in high dose group had statistically significant (P<0.05 or P<0.01), while the indicators of low dose group showed trendy of improvement. Compared with the sham surgery group, the t-PA and t-PA/PAI-I of the model group were significantly reduced(P<0.01), but there was no significant difference in PAI-I. Compared with the model group, the t-PA of the high and low dose groups and the t-PA/PAI-I of high dose group were obviously increased(P<0.05 or P<0.01). There was no significantly difference in PAI-1. Compared with the model group, the dry weight and wet weight of thrombus decreased in SNFE groups, and the inhibition rates were 56.86% and 41.67% in high dose group and 21.57% and 11.90% in low dose group respectively. Dense thrombus was found in the blood lumen of the model group, the low dose group had less thrombus than the model group, and there was no obvious thrombus in the aspirin group and the high dose group. CONCLUSION SNFE can inhibit the formation of common carotid artery thrombosis induced by FeCl3 in rats by affecting coagulation and fibrinolysis systems.
