糖尿病及其并发症是日益严重的代谢流行疾病,是世界范围内一个重要的健康和社会经济问题。因此,尚需寻找新的潜在药物靶点,从而减少糖尿病和相关并发症的发生发展。非甾体抗炎药激活因子基因-1 (non-steroidal anti-inflammatory drug activated gene-1, NAG-1),又称生长分化因子15 (growth differentiation factor 15,GDF15),是转化生长因子-β(transforming growth factor-β,TGF-β)超家族的分化成员(以下简称NAG-1/GDF15)。NAG-1/GDF15在多种疾病中发挥重要的调节作用,包括癌症、心血管疾病、肥胖和糖尿病等。本文简要介绍了NAG-1/GDF15的结构及表达调控机制,重点介绍了NAG-1/GDF15在糖尿病和糖尿病肾病中的研究进展和分子机制,并对靶向NAG-1/GDF15抗糖尿病药物干预研究进行总结。本综述旨在为NAG-1/GDF15在糖尿病及相关代谢疾病中的基础研究和靶向NAG-1/GDF15抗糖尿病药物开发提供理论依据。 |

Diabetes mellitus and its complications are serious epidemic diseases associated with metabolism and an important health and socioeconomic problem worldwide. Therefore, there is still a need to find new drug targets to reduce the occurrence and development of diabetes and related complications. Non-steroidal anti-inflammatory drug activator gene-1, NAG-1, also known as growth differentiation factor 15 (GDF15) is a divergent member of the transforming growth factor-β (TGF-β) superfamily (hereinafter referred to as NAG-1/GDF15). NAG-1/GDF15 plays an important regulatory role in a variety of diseases, including cancer, cardiovascular disease, obesity, and diabetes. This review briefly introduces the structure and expression regulation mechanism of NAG-1/GDF15, focuses on the research progress and molecular mechanism of NAG-1/GDF15 in diabetes mellitus and diabetic nephropathy, and summarizes the research of anti-diabetic drug intervention targeting NAG-1/GDF15. The aim is to provide a theoretical basis for the basic research of NAG-1/GDF15 in diabetes mellitus and related metabolic diseases and the development of anti-diabetic drugs targeting NAG-1/GDF15. |
