摘要: |
目的:拟采用UPLC-Q-Exactive-Orbitrap技术对白豆蔻水提物主要成分及大鼠口服入血成分进行分析。方法:运用ACQUITY UPLC HSS T3色谱柱(2.1×100 mm,1.8 μm);以A(去离子水,含0.1%甲酸)和B(乙腈,含0.1%甲酸)为流动相,梯度洗脱,柱温:35℃;进样体积:10 μL;流速:0.2 mL/min。采用电喷雾离子源(ESI),Full ms/dd-ms2模式检测。结果:从白豆蔻水提物中共鉴定70种成分,包括有机酸类、黄酮类、氨基酸类、生物碱类、内酯类等。含药血清样本分析出19种原型成分,包括青蒿酸、壬二酸等11个有机酸类、异土木香内酯、去氢木香内酯等2种内酯类、甜菜碱、黄嘌呤等2种生物碱类、酪氨酸、色氨酸等3种氨基酸和1种其他类成分。结论:本研究首次阐明了白豆蔻水提物化学组成及口服入血成分,为其进一步研究和开发提供理论依据。 |

OBJECTIVE: UPLC-Q-Exactive-Orbitrap technology is proposed to analyze the main components of Baidoukou aqueous extract and the components that enter the blood of rats after oral administration. METHODS: Using ACQUITY UPLC HSS T3 column (2.1×100 mm, 1.8 μm); Using A (deionized water, containing 0.1% formic acid) and B (acetonitrile, containing 0.1% formic acid) as mobile phases, gradient elution, column temperature: 35°C; injection volume: 10 μL; flow rate: 0.2 mL/min. Using electrospray ionization source (ESI), detect with Full ms/dd-ms2 mode. RESULTS: A total of 70 components were identified from the aqueous extract of Baidoukou, including organic acids, flavonoids, amino acids, alkaloids, and lactones. 19 prototype components were identified in the drug-containing serum samples, including 11 organic acids such as artemisinic acid and azelaic acid, 2 lactones such as isoalantolactone and dehydrocostus lactone, 2 alkaloids such as Betaine and Xanthine, 3 amino acids such as L-Tyrosine and L-Tryptophan, and 1 other component.CONCLUSIONS: It"s clarified for the first time the chemical composition of Baidoukou aqueous extract and its oral blood components in this study, providing a theoretical basis for its further research and development. |
