基于UPLC-Q-TOF-MS/MS的银线莲化学成分分析

摘要:
目的 采用超高效液相色谱-四极杆飞行时间质谱联用技术(UPLC-Q-TOF-MS/MS)快速鉴定银线莲中的化学成分。方法 采用Cortecs UPLC C18(2.1×100mm,1.6μm)色谱柱,流动相为甲醇-0.1%甲酸水,梯度洗脱,流速为0.25 mL/min,柱温45℃。飞行时间质谱采用正负离子模式扫描。结果 根据UPLC-Q-TOF-MS/MS法推断鉴定出银线莲中28个化学成分,包括黄酮类成分7个,酚类成分15个,2-异丁基苹果酸葡萄糖氧基苄酯类成分4个,其他苷类成分2个。结论 UPLC-Q-TOF-MS/MS技术为鉴定银线莲中化学成分提供了一种快速、高效的定性分析方法,研究结果可为银线莲的物质基础及质量标准研究提供理论依据。

Analysis of chemical constituents of Goodyerae Schlechtendalianae Herba based on UPLC-Q-TOF-MS/MS

Abstract:
OBJECTIVE To identify the chemical constituents in Goodyerae Schlechtendalianae Herba by UPLC-Q-TOF-MS/MS. METHODS Cortecs UPLC C18 (2.1×100 mm, 1.6 μm ) column was used. The mobile phase was methanol-0.1 %formic acid with gradient elution. The flow rate was 0.25 mL/min and the column temperature was 45℃. MS monitoring was performed by electrospray ionization in positive and negative ion switching mode. RESULTS 28 chemical constituents in Goodyerae Schlechtendalianae Herba were identified, including 7 flavonoids, 15 phenols, 4 2-isopropyl-(glucosyloxy) benzyl malat and 2 other glycosides. CONCLUSION UPLC-Q-TOF-MS/MS provides a rapid and efficient qualitative analysis method for the identification of chemical constituents in Goodyerae Schlechtendalianae Herba. The research results can provide a theoretical basis for the material basis and quality standard research of Goodyerae Schlechtendalianae Herba.
