六神曲成分的快速鉴定及脂类活性研究

摘要:
目的:快速鉴定六神曲的成分,并探索其脂类活性,为六神曲发酵工艺改进与质量提升提供依据。方法:采用HPLC-Q-TOF-MS与GNPS分子网络分析结合的技术,通过Lipidomics、HMBD、METLIN等数据库匹配,依据文献数据、质谱裂解规律并结合GNPS分子网络分析,鉴定六神曲的化学成分,用细胞炎症实验评价六神曲脂类物质的抗炎作用。结果:从六神曲中共鉴定出72个化学成分,包括黄酮、倍半萜、鞘脂、甘油糖酯及取代苯类成分,脂类成分具有抗炎活性。结论:从六神曲中发现的磷脂酰胆碱、磷脂酰乙醇胺、双半乳糖甘油酯、单萜等10类成分以往未见报道,鉴定的72个成分中除化合物8、9、11~15、21、22外其余63个成分为首次从六神曲中鉴定,为六神曲的发酵机理与活性成分研究提供了基础数据。

Abstract:
Objective: To elucidate the chemical constituents of Massa Medicata Fermentata (MMF) for improvement of fermentation process and quality evaluation. Methods: HPLC-Q-TOF-MS and GNPS molecular network were performed to analyze the main chemical constituents of MMF. The chemical constituents were identified by automatically matching HMBD, METLIN, Lipidomics and compared with the data reported in the literature, and mass fragmentation characteristics analysis, as well as the correlation of known and unknown nodes in the GNPS molecular network. Results: Seventy-two compounds were identified, including Sphingolipids, Glycerolipids, flavone, sesquiterpene and aromatic compound. Lipid components from MMF express anti-inflammatory activity. Conclusion: Ten structural classification and 63 compounds except compound 8, 9, 11~15, 21, 22 were identified for the first time from MMF. This result provides the basic data for the study of fermentation mechanism and active components of MMF
