化妆品中10株洋葱伯克霍尔德菌的筛查鉴定及耐药基因研究

摘要:目的 对2017~2022年化妆品国家级监督抽检、浙江省本级监督抽检和行政许可检验中分离的149株污染微生物,进行洋葱伯克霍尔德菌的筛查鉴定,并针对洋葱伯克霍尔德菌开展耐药基因研究。方法 采用液体增菌法和洋葱伯克霍尔德菌选择性琼脂培养基(Burkholderia Cepacia Selective Agar,BCSA)平板划线法对2017~2022年收集的149株化妆品污染微生物进行复苏、分离和筛查。针对BCSA上可疑洋葱伯克霍尔德菌菌落,采用特异性洋葱伯克霍尔德菌群引物和探针,进行实时荧光定量PCR(Quantitative Real-time PCR,qPCR)鉴定,后采用16S rDNA测序技术确证鉴定,根据鉴定结果,进一步采用二代测序技术针对洋葱伯克霍尔德菌进行耐药基因研究。结果 本研究从149株化妆品污染微生物中共鉴定到10株洋葱伯克霍尔德菌,均隶属于(Burkholderia cepacia complex,Bcc),二代测序检测到耐药基因26种,提示该菌可能对青霉素、多粘菌素和大环内酯类抗生素等约33种抗生素耐药,检测到多重耐药基因11种,耐药机理多与产生外排泵有关,提示化妆品中分离的洋葱伯克霍尔德菌存在一定的多重耐药安全隐患。结论 149株化妆品污染微生物中鉴定到10株洋葱伯克霍尔德菌,耐药基因分析,该菌具有一定的多重耐药安全隐患,应考虑经风险评估后将该菌纳入微生物检测指标。

Abstract:
ABSTRACT:OBJECTIVE To screen and identify Burkholderia cepacia from 149 strains of contaminated microorganisms isolated from national supervision and sampling of cosmetics, Zhejiang provincial supervision and administrative licensing during 2017-2022, At the same time, the resistance genes of Burkholderia cepacia were studied. METHODS A total of 149 strains of bacteria in cosmetics collected from 2017 to 2022 were resuscitated, isolated and screened by liquid enrichment method and Burkholderia Cepacia Selective Agar ( BCSA ) plate streaking method. Quantitative real-time PCR (qPCR) was used to identify suspicious Burkholderia cepacia colonies on BCSA using specific Burkholderia cepacia complex primers and probes, further identification was carried out by using 16S rDNA sequencing technology. According to the identification results, the second generation sequencing technology was used to study the resistance genes of Burkholderia cepacia. RESULTS In this study, 10 strains of Burkholderia cepacia were identified from 149 strains of bacteria contaminated by cosmetics. All of them belonged to Burkholderia cepacia complex, the results suggested that the bacteria might be resistant to penicillin, Polymyxin and Macrolide, and 11 multi-drug resistance genes were detected. It is suggested that Burkholderia cepacia isolated from cosmetic products has some hidden danger of multi-drug resistance. CONCLUSION A total of 10 strains of Burkholderia cepacia were identified from 149 strains of microorganisms contaminated in cosmetics. The analysis of drug-resistant genes indicated that the bacteria had potential safety risks. It should be considered that the bacteria should be included in the microbial detection index after risk assessment.
