基于UHPLC-LTQ-Orbitrap-MS/MS技术快速分析六神曲中的化学成分

摘要:
目的 建立快速,高效的UHPLC-LTQ-Orbitrap-MS/MS 方法,系统,全面地对六神曲化学成分进行快速的定性分析。为六神曲质量标准方法的开发,质量控制方法的完善以及临床应用提供一定的理论依据。方法 采用超高效液相色谱-线性离子阱轨道阱串联质谱联用技术(UHPLC-LTQ-Orbitrap-MS/MS)扫描六神曲中化合物的精确分子量,经对照品和多级碎片离子信息,并结合相关文献以及裂解规律,从而对六神曲实现准确的定性分析。结果 共鉴定出76种化学成分,包括氨基酸类、核苷类、黄酮类、有机酸类、苯丙素类、萜类、氰苷类、不饱和脂肪酸类等。 结论 建立了对六神曲化学成分的快速定性分析的方法,本研究为六神曲的质量评价指标及药效物质基础研究提供了一定的理论依据。

Abstract:
ABSTRACT:OBJECTIVE To establish an efficient and stable uhplc-ltq-orbitrap-ms / MS method for systematic and comprehensive qualitative analysis of chemical components in Massa Medicata Fermentata. To provide basis for the development of quality control methods, improvement of standards and clinical application of Massa Medicata Fermentata.METHODS Ultra high performance liquid chromatography-linear ion trap orbital trap tandem mass spectrometry (UHPLC-LTQ-Orbitrap-MS/MS) was used to scan the precise molecular weight of the compounds in Massa Medicata Fermentata, and accurate qualitative analysis of Massa Medicata Fermentata was achieved through reference substance and multi-stage fragment ion information, combined with relevant literature and fragmentation rules. RESULTS A total of 76 chemical constituents were identified, including amino acids, nucleosides, flavonoids, organic acids, phenylpropanoids, terpenoids, cyanosides, unsaturated fatty acids and the others. CONCLUSION A rapid qualitative analysis method for the chemical components of Massa Medicata Fermentata was established. This study provides a theoretical basis for the quality evaluation index and the material basis of drug effect of Massa Medicata Fermentata.
