摘要: |
目的 建立辅酶A中残留溶剂丙酮、巯基乙醇的含量测定方法。方法 采用顶空毛细管气相色谱法,HP-INNOWAX毛细管柱(30 m×0.53 mm×1 μm),FID检测器。结果 丙酮、巯基乙醇达到了完全分离,浓度与峰面积呈良好的线性关系(r>0.999),平均回收率分别为101.29%(RSD=1.32%,n=9)、98.67%(RSD=1.71%,n=9),检测限分别为2.52 μg·mL-1、3.95 μg·mL-1,样品中丙酮含量为0.06%~4.33%,巯基乙醇含量为0.14%~0.72%。结论 本方法快速准确可靠,简便易行,可用于辅酶A的质量控制。 |

Abstract: |
OBJECTIVE To establish a headspace capillary gas chromatography for determining the residual organic solvents acetone and thioglycol in coenzyme A. METHODS Using a HP-INNOWAX capillary column (30 m×0.53 mm×1 μm), flame ionization detector (FID) and adopting headspace injection. RESULTS The calibration curves of each solvent had good linear relationship(r>0.999), The average recoveries was 101.29% (RSD=1.32%, n=9), 98.67% (RSD=1.71%, n=9), and the detection limit was 2.52 μg·mL-1, 3.95 μg·mL-1. The content of acetone ranged from 0.06% to 4.33%,and the content of thioglycol ranged from 0.14% to 0.72%. CONCLUSION The proposed method offers simple and fast operation, high sensitivity and accuracy, and it is a substitute method to determinate the residual organic solvents in coenzyme A. |
