盐酸小檗碱对B16细胞的分化诱导作用

摘要:
目的 测定盐酸小檗碱对小鼠黑色素瘤B16细胞的诱导分化作用。方法 以MTT法测定盐酸小檗碱对B16细胞增殖的抑制作用,通过克隆形成实验、细胞黑色素含量的测定以及B16细胞体内成瘤能力的测定观察盐酸小檗碱对B16细胞的诱导分化作用。结果 盐酸小檗碱对B16细胞具有明显的增殖抑制作用;并使B16细胞生长缓慢,平铺不重叠,呈正常上皮样细胞分化,细胞的克隆形成能力降低,细胞内的黑色素生成能力增强,C57/BL小鼠体内成瘤能力降低。结论 盐酸小檗碱对B16细胞具有诱导分化作用。

Abstract:
OBJECTIVE To investigate the inducing differentiation effect of berberine hydrochloride on B16 mouse melanoma cells. METHODS MTT assay was used to evaluate the inhibitory effect of berberine hydrochloride on proliferation of B16 cells. Morphological changes, melanin content, cloning efficiency and tumorigenicty in C57/BL mice were also used to estimate the differentiation inducing activity of berberine hydrochloride in B16 cells. RESULTS Berberine hydrochloride significantly inhibited the proliferation of B16 cells. After treatment with berberine hydrochloride, B16 cells differentiated into mature epithelia-like cells, cloning efficiency of B16 cells decreased, the content of melanineon increased and tumorigenicty in C57 mice decreased. CONCLUSION Berberine hydrochloride can induce differentiation of B16 cells.
