HPLC-ELSD测定三黄合剂中黄芪甲苷的含量

摘要:
目的 建立三黄合剂中黄芪甲苷含量测定的HPLC方法并制定该制剂的质量控制指标。方法 采用Diamonsil C18柱,流动相为乙腈-水(35∶65),流速:1.0 mL·min-1;漂移管温度为95 ℃,氮气流速为3.0 L·min-1。结果 进样量在0.25~5.00 μg内,黄芪甲苷浓度的对数值lgC与峰面积积分的对数值lgA之间呈良好的线性关系,r=0.999 3。结论 本法操作简便,准确,可用于三黄合剂的质量控制。

Abstract:
OBJECTIVE To establish a method using HPLC coupled with ELSD for determination of astragaloside IV in Sanhuang mixture and formulate a standard for qulity control. METHODS A Diamonsil C18 column(200 mm×4.6 mm, 5 μm) was used, acetonitrile-water (35∶65) was used as mobile phase and the flow rate was 1.0 mL·min-1. The temperature of drift tube for ELSD was 95 ℃ and the flow rate of nitrogen was 3.0 L·min-1. RESULTS The linear range for astragaloside IV was 0.25-5.00 μg (r=0.999 3), the RSDs of intra- and inter-day were less than 2.0%. CONCLUSION The method is simple, accurate and suitable for quality control of Sanhuang mixture.
