HPLC比较不同产地生药八角莲中鬼臼类成分含量

摘要:
目的 比较不同产地的八角莲中鬼臼毒素和4’-去甲鬼臼毒素含量。方法 采用RP-HPLC,色谱柱C18(250 mm×4.6 mm,5 μm),流动相为甲醇-0.1 mol·L-1K2HPO4(50∶50),流速1.0 mL·min-1,检测波长207 nm,柱温25 ℃。结果 鬼臼毒素的线性范围20~400 ng,r=0.999 9,回收率100.4%,RSD=1.2%;4’-去甲鬼臼毒素的线性范围2~20 ng,r=0.999 8,回收率101.6%,RSD=2.5%。结论 该方法准确、可靠、简便,不同地域的生药八角莲中鬼臼类成分含量差异明显

Abstract:
OBJECTIVE To compare the contents of podophyllotoxin and 4'-demethylpodophyllotoxin in Rhizoma et Radix Dysosmae of different areas. METHODS The assay was performed on phenomenex C18(250 mm×4.6 mm, 5 μm). The mobile phase was MeOH-0.1 mol·L-1K2HPO4(50∶50). Flow rate was 1.0 mL·min-1. Wavelength was 207 nm, and the temperature of column was 25 ℃. RESULTS The calibration curve was linear in the range of 20-400 ng for podophyllotoxin (r=0.999 9). The average recovery was 100.4% (RSD=1.2%). And the liner range of demethylpodophyllotoxin was 2-20 ng (r=0.999 8). The recovery was 101.6%(RSD=2.5%). CONCLUSION The method is accurate, reliable and simple, the apparent differences of contents among samples was found.
