双波长HPLC同时测定熊胆丸中栀子苷和黄芩苷的含量

摘要:
目的 建立HPLC同时测定熊胆丸中栀子苷和黄芩苷含量的方法。方法 色谱柱为 Hibar C18柱(150 mm×4.6 mm,5 μm);流动相:A相为0.2%磷酸水溶液,B相为乙腈;梯度洗脱,B:15%(0~9 min),15%→25% (9~10 min),25% (10~17 min );流速为 1.0 mL·min-1;柱温为室温;检测波长分别为240 nm(栀子苷),278 nm(黄芩苷)。结果 栀子苷、黄芩苷保留时间分别为4.5,15.6 min左右,进样质量分别在0.08~0.8,0.23~2.3 μg内线性关系良好,线性回归方程分别为Y=1 413.8X+20.75,r=0.999 8(n=5);Y=2 788.7X+11.4,r=0.999 9(n=5),平均回收率分别为96.0%,103.6%,RSD分别为1.75%,2.01%。结论 该法快速、准确,重复性、精密度良好,适用于测定熊胆丸中栀子苷和黄芩苷的含量。

Abstract:
OBJECTIVE To establish a double wavelength HPLC for simultaneously determination of geniposide and baicalin in Xiongdan pills. METHODS The analysis was performed on an Hibar C18 column (150 mm×4.6 mm, 5 μm) with gradient elution using 0.2% phosphoric acid (A) and acetoniterile (B) in the order of B 15%(0-9 min), 15%→25%(9-10 min), 25%(10-17 min), the flow rate was 1.0 mL·min-1. Geniposide and baicalin were detected at 240 nm and 278 nm, respectively. The column temperature was room temperature. RESULTS The calibration curves of geniposide and baicalin showed good linearity in the ranges of 0.08-0.8 μg and 0.23-2.3 μg. The average recoveries were 96.0% and 103.6% with RSD of 1.75% and 2.01%. CONCLUSION The method is simple, accurate and reproducible for simultaneous determining geniposide and baicalin in Xiongdan Pills.
