戒毒康酊剂中氢溴酸东莨菪碱及总生物碱的含量测定

摘要:
目的 测定中药戒毒康酊剂中氢溴酸东莨菪碱及总生物碱的含量。方法 采用Diamonsil C18柱 (250 mm×4.6 mm, 5 μm)分离测定氢溴酸东莨菪碱,流动相:以甲醇-0.02 mol·L-1醋酸钠溶液(含0.02%三乙胺和0.3%四氢呋喃,用冰醋酸调pH到6.2)梯度洗脱;流速1 mL·min-1;检测波长:206 nm。采用酸性染料比色法测定戒毒康中的总生物碱,0.2%溴百里香酚兰溶液显色,420 nm处测得吸光度。结果 氢溴酸东莨菪碱在1.0~5.0 μg内呈良好的线性关系(r=0.999 6);平均回收率为98.8%,RSD为2.29%(n=6)。总生物碱在0.04~0.2 mg内呈良好的线性关系(r=0.999 8);平均回收率为97.6%,RSD为2.06%(n=6)。结论 本方法简便、快速、准确、专属性强,可作为戒毒康酊剂的检验方法。

Abstract:
OBJECTIVE To establish an HPLC method for determination of contents of scopolamine hydrobromide and total alkaloid in Jiedukang tincture. METHODS A Diamonsil C18 column(250 mm×4.6 mm, 5 μm) was used to isolate and determine scopolamine hydrobromide. The mobile phase was methanol-0.02 mol·L-1 NaAc solution (with 0.02% triethylamine and 0.3% tetrahydrofuran, adjusted to pH 6.2 by HAc). The flow rate was 1 mL·min-1. The detection wavelength was set at 206 nm. Total alkaloids in Jiedukang tincture were determined with acid dye colorimetry. 0.2% Bromothymol blue was used as the dye reagent and the wavelength was set at 420 nm. RESULTS The linear range of scopolamine hydrobromide was 1.0-5.0 μg with a good correlation (r=0.999 6). The average recoveries was 98.8% with RSD of 2.29%(n=6). The total alkaloids showed good linearity within the range of 0.04-0.2 mg(r=0.999 8), the average recoveries was 97.6% with RSD of 2.06%(n=6). CONCLUSION The method established is sensitive, accurate and can be used for determination of scopolamine hydrobromide in Jiedukang tincture.
